The filing represents another step in GenBio’s broader strategy of developing and protecting intellectual property associated with its emerging scientific programs as research progresses.

BlockR™ is being developed from a body of scientific investigation extending over approximately 15 years into the biological activity of selected nature-derived extracts, their constituent molecules and potential mechanisms relevant to inflammation, immune response and other therapeutic applications.

During the COVID-19 pandemic, scientific interest increased around the potential biological activity of selected Australian native plant extracts and the mechanisms through which naturally occurring compounds might influence inflammatory and immune-response pathways.

Research associated with the scientific foundation underlying GenBio’s program has subsequently examined mechanisms involving nuclear factor kappa B (NF-κB), an important regulator of inflammatory and immune signaling.

Published laboratory research involving selected Australian botanical extracts has reported modulation of inflammatory mediators and NF-κB-related activity, providing further scientific rationale for investigating the underlying bioactive compounds and their potential mechanisms.

GenBio emphasizes that laboratory findings involving inflammatory pathways do not establish prevention or treatment of COVID-19 or any other disease in humans. Rather, the research contributed to a broader scientific foundation supporting continued investigation of the biological mechanisms involved.

Subsequent phytochemical investigation has sought to identify individual molecules contributing to observed biological activity. This research has also identified mechanisms of interest to GenBio’s broader scientific programs, including efflux pump inhibition, which the Company continues to evaluate as part of its therapeutic research strategy.

Development Focus Expands Toward Gastrointestinal Applications

As the BlockR™ program progresses, GenBio anticipates increasing its research focus on potential gastrointestinal applications and pathogens.

The Company is evaluating a formulation strategy intended to protect selected bioactive components from the acidic environment of the stomach and provide targeted release primarily within the gastrointestinal tract.

This approach remains under development and will require further formulation, pharmacological, toxicological and efficacy research before its potential utility can be determined.

An important planned stage of the program involves further refinement of the underlying extract, including evaluation of whether an inactive class of molecules can be removed based upon prior scientific research.

The objective is to determine whether a more concentrated and scientifically defined composition can be developed while retaining or improving the biological activity identified during earlier research.

From Laboratory Activity to Biological Validation

GenBio believes one of the most important challenges facing nature-derived therapeutic development is translation.

Compounds demonstrating promising activity in laboratory models do not necessarily achieve sufficient exposure, stability, bioavailability or biological activity when administered to a living system.

Accordingly, the Company intends for future development of BlockR™ to include rigorous evaluation of bioavailability, absorption, distribution, metabolism and excretion (ADME), drug metabolism and pharmacokinetics (DMPK), and toxicity, including appropriate in vivo studies.

These studies will be intended to determine whether the scientific activity identified during earlier stages of research can support continued development toward potential human applications.
